Immunoglobulins is a group of glycosylated proteins secreted by plasma cells which acts as antibodies in the immune response against specific antigens. There are five classes of immunoglobulins: IgA, IgD, IgE, IgG, and IgM.

The production of immunoglobulins are done by the hybridoma technology which is reviewd at production of immunoglobulins or immunoglobulin production.

Key requirements for successful immunoglobulin production include immunization strategies, antigen design, and screening technologies.

 

Antigen design and production

It is important to have a purified antigen to use for the immunoglobulins production. We provide service of protein production in different expression systems including E. coli, yeast, insect or mammalian cell. Alternatively, we offer peptide synthesis and conjugation.

 

Immunizations

We immunize mice with in-house protocols that involve proper adjuvants, inoculation routes, dosage and timing. We use different formats of immunogens for animal immunizations depending on the specific purpose.  We immunize 5 mice per antigen and perform sera testing by ELISA.

 

Hybridoma

We fuse splenocytes with proper myeloma cells using optimized PEG mediated fusion protocols. We screen hybridoma clones with customized protocols according to the final applications.  In this step of the immunoglobulin production, we perfrorm fusion, colony screening, subcloning, colony expansion and freezing of positive clones.

 

Ascites Production and immunoglobulin purification 

Inoculation of hybridoma cells in to relevant animals to produce ascites. Immunoglobulin purification from ascites by protein A/G (2 clones, 3mg each). In addition, we can perform large scale immuniglobulin production since we have an antibody fermentation capacity of over 300L.
